Clearazide:
Clearazide
is a topically delivered cytotoxic agent that is currently in pivotal
clinical studies for treating cutaneous T-cell lymphoma (CTCL). Clearazide
works by suppressing T-cell proliferation thereby reducing or clearing
skin lesions associated with CTCL. Clearazide has also demonstrated
activity in other T-cell proliferating diseases such as Psoriasis and
Alopecia Areata. In CTCL, Clearazide has been shown to be effective and
safe in multiple clinical studies in hundreds of CTCL patients. Yaupon
is currently working with 13 of the top dermatological cancer centers
in the US and has received an FDA grant to support clinical activities.
Upon FDA approval, Clearazide will be the first topical cytotoxic agent
approved for treating this orphan disease, which affects roughly 50,000
in the US and EU.
Lobeline
Lobeline
is a dopamine modulating agent for the treatment of methamphetamine
addiction and Attention Deficit Hyperactivity Disorder. Lobeline is
currently in phase 2 clinical testing for both indications. Lobeline
works through a unique dual mechanism of action. In one pathway it
binds to nicotinic receptors and through another pathway it binds to
VMAT2 receptors. Since both of these receptor targets are part of the
dopaminergic cascade, Lobeline has the ability to modulate overactive
dopamine production caused by disease or through the use of drugs of
abuse. Yaupon is working through a partnership with the NIH to develop
lobeline as the first pharmacological treatment for methamphetamine
addiction, which is the fastest growing drug of abuse in the US and it
affects over one million Americans.
YT-1006
YT-1006 is an NMDA antagonist
for the treatment of neuropathic pain that eliminates classic NMDA side-effects
through a unique stereochemistry approach. NMDA antagonists are well
known to be very good analgesics, but their clinical utility is limited
due to dose related side-effects such as hallucinations and sedation.
YT-1006, on the other hand, is a unique NMDA antagonist in that has
been shown to have a wide therapeutic index (dose related side effects
occur well above the therapeutic dose range). In animal models of
neuropathic pain YT-1006 demonstrated very good analgesic activity,
while toxicological experiments exhibiting no observed side-effects in
the efficacy dose range. YT-1006 is orally active and also reduces
opiate tolerance. Yaupon has received significant funding from the NIH
through a competitive SBIR grant that will lead to a phase 1 proof of
concept study in 2009.
Other
Products
Yaupon
has other product programs in the pipeline focused on dermatological
and Central Nervous System diseases. One program is a topically
delivered plant based extract that is being study for various skin inflammatory
conditions. This product has a unique mechanism of action that
interrupts the inflammatory cascade. Yaupon expects to file an IND for
this drug substance in 2009. Other programs include efforts in Tourette
Syndrome, movement disorders, addiction and mental illness.
